This study was aimed at the development of a conductometric biosensor based on acetylcholinesterase considering the feasibility of its application for the inhibitory analysis of various toxicants. In this paper, the optimum conditions for enzyme immobilization on the transducer surface are selected as well as the optimum concentration of substrate for inhibitory analysis. Sensitivity of the developed biosensor to different classes of toxic compounds (organophosphorus pesticides, heavy metal ions, surfactants, aflatoxin, glycoalkaloids) was tested. It is shown that the developed biosensor can be successfully used for the analysis of pesticides and mycotoxins, as well as for determination of total toxicity of the samples. A new method of biosensor analysis of toxic substances of different classes in complex multicomponent aqueous samples is proposed. 相似文献

A liquid chromatography‐tandem mass spectrometry (LC‐MS/MS) method was developed and validated for the simultaneous determination of 12 constituents of Gegen Qinlian Decoction (GQD), namely puerarin, daidzein, baicalin, wogonoside, wogonin, liquiritin, liquiritigenin, berberine, jatrorrhizine, palmatine, coptisine and glycyrrhetic acid, in rat plasma. The plasma samples were spiked with the internal standard (IS) carbamazepine acidified with HCl and extracted by liquid–liquid extraction with ethyl acetate. Chromatographic separation was achieved on a Shiseido Capcell PAK C18 column utilizing a gradient elution profile and a mobile phase consisting of (A) 0.1% formic acid in water and (B) acetonitrile. Detection was performed in the multiple reaction monitoring mode using electrospray ionization in the positive ion mode at a flow rate of 0.3 mL/min and a run time of 8 min. All of the calibration curves gave good linearity (r > 0.9930) over the concentration range from 0.6–360 to 16.2–9720 ng/mL for all components. The intra‐ and inter‐day precisions were <15.0% in terms of the relative standard deviation, and the accuracies were within ±13.7% in terms of the relative error. The method was successfully applied to investigate the pharmacokinetics of the major active compounds of Gegen Qinlian Decoction after its oral administration to rats. Fast and effective structural/compositional analysis on formulated systems represents one of the major challenges encountered in analytical science. 13C‐detected diffusion represents a promising tool to tackle the aforementioned challenges, particularly in industry. Toward exploring the generic applications of 13C‐detected diffusion, thermal convection induced by 1H decoupling has been identified as a key factor that resulted in significantly reduced resolution in the diffusion dimension. Optimization of experimental parameters and utilization of double‐stimulated echo‐based pulse sequence both can effectively suppress the thermal convection caused by the 1H decoupling, the success of which allows robust and generic applications of 13C‐detected diffusion to systems from mixtures of small molecules, polymer blends, and copolymers to actual complex formulated systems. The amount of data collected during synchrotron X-ray diffraction (XRD) experiments is constantly increasing. Most of the time, the data are collected with image detectors, which necessitates the use of image reduction/integration routines to extract structural information from measured XRD patterns. This step turns out to be a bottleneck in the data processing procedure due to a lack of suitable software packages. In particular, fast-running synchrotron experiments require online data reduction and analysis in real time so that experimental parameters can be adjusted interactively. Dioptas is a Python-based program for on-the-fly data processing and exploration of two-dimensional X-ray diffraction area detector data, specifically designed for the large amount of data collected at XRD beamlines at synchrotrons. Its fast data reduction algorithm and graphical data exploration capabilities make it ideal for online data processing during XRD experiments and batch post-processing of large numbers of images. 相似文献

For the first time, the influence of different types of atoms (Zn and O) on the antibacterial activities of nanosized ZnO was quantitatively evaluated with the aid of a 3D‐printing‐manufactured evaluation system. Two different outermost atomic layers were manufactured separately by using an ALD (atomic layer deposition) method. Interestingly, we found that each outermost atomic layer exhibited certain differences against gram‐positive or gram‐negative bacterial species. Zinc atoms as outermost layer (ZnO?Zn) showed a more pronounced antibacterial effect towards gram‐negative E. coli (Escherichia coli), whereas oxygen atoms (ZnO?O) showed a stronger antibacterial activity against gram‐positive S. aureus (Staphylococcus aureus). A possible antibacterial mechanism has been comprehensively discussed from different perspectives, including Zn2+ concentrations, oxygen vacancies, photocatalytic activities and the DNA structural characteristics of different bacterial species. 相似文献

The steady flow and heat transfer of a hybrid nanofluid past a permeable stretching/shrinking wedge with magnetic field and radiation effects are studied. The governing equations of the hybrid nanofluid are converted to the similarity equations by techniques of the similarity transformation. The bvp4c function that is available in MATLAB software is utilized for solving the similarity equations numerically. The numerical results are obtained for selected different values of parameters. The results discover that two solutions exist, up to a certain value of the stretching/shrinking and suction strengths. The critical value in which the solution is in existence decreases as nanoparticle volume fractions for copper and wedge angle parameter increase. It is also found that the hybrid nanofluid enhances the heat transfer rate compared with the regular nanofluid. The reduction of the heat transfer rate is observed with the increase in radiation parameter. The temporal stability analysis is performed to analyze the stability of the dual solutions, and it is revealed that only one of them is stable and physically reliable. 相似文献

A new non-perturbative approach is proposed to solve time-independent Schr?dinger equations in quantum mechanics.It is based on the homotopy analysis method(HAM)that was developed by the author in 1992 for highly nonlinear equations and has been widely applied in many fields.Unlike perturbative methods,this HAM-based approach has nothing to do with small/large physical parameters.Besides,convergent series solution can be obtained even if the disturbance is far from the known status.A nonlinear harmonic oscillator is used as an example to illustrate the validity of this approach for disturbances that might be one thousand times larger than the possible superior limit of the perturbative approach.This HAM-based approach could provide us rigorous theoretical results in quantum mechanics,which can be directly compared with experimental data.Obviously,this is of great benefit not only for improving the accuracy of experimental measurements but also for validating physical theories. 相似文献

噪声免疫腔增强光外差分子光谱技术(NICE-OHMS)是目前世界上最灵敏的激光吸收光谱技术,其在低压环境中具有极高的探测灵敏度。然而当测量样品处于大气压时,NICE-OHMS系统的探测灵敏度会大幅下降。主要原因之一是大气压下获取最大NICE-OHMS信号幅度的条件与低气压下不同。通过对大气压NICE-OHMS理论进行分析,分析了影响信号幅度的参数,并通过数值模拟来寻找最佳的实验条件。本文着重讨论影响信号的主要参数包括光学腔腔长L,调制系数β,探测相位θ。其中,由于在NICE-OHMS中使用DeVoe-Brewer技术将调制频率ν_m锁定到Fabry-Parot(FP)腔的自由光谱区(FSR)。因此FP腔的腔长决定了ν_m,同时还作用于信号幅度S■。模拟结果显示,当腔长增大时,由于ν_m随之减小,载波和边带的光谱成分相互重叠部分增大,因此线型函数的幅度逐渐减小。而吸收信号幅度随着腔长的增加而逐渐增加,色散信号幅度先增大后减小,并且在腔长等于8 cm时达到最大值。调制系数β会影响频率调制后激光载波和边带的幅度大小,并且影响信号线型。随着腔长的增加,最大信号幅度对应的β值也随之增加。在相同腔长下,色散信号的最佳β值小于吸收信号,更容易使用电光调制器实现。最后分析了参数的可实现性,分析了不同种类激光器的频率调谐能力,压电陶瓷的扫描宽度等。以乙炔气体为例,大气压下NICE-OHMS的谱线半宽达到~3 GHz,而光谱覆盖范围大于10 GHz。分布反馈式半导体激光器(DFB)与外腔二极管激光器(ECDL)的频率调谐范围可以达到30 GHz以上,但是由于激光线宽宽,得到的PDH锁定性能欠佳。回音壁模式激光器(WGM)和掺饵光纤激光器(EDFL)线宽为百Hz量级,是目前高灵敏NICE-OHMS系统中常用的光源。但是WGM目前可以实现了5 GHz的激光频率调谐范围,而EDFL的外部电压可控制的调谐范围仅为3 GHz。使用精细度为55000的腔进行模拟,调制系数β=1,腔长大于8 cm时,可使用WGM激光器实现,腔长大于25 cm时,可以使用EDFL激光器实现。而对于在设计光学腔中常用的伸缩长度为25μm的PZT,随着腔长的增加,对应的腔模频移范围逐渐减小,在腔长为典型的40 cm时,扫描范围大于12 GHz。 相似文献
